Title: Development of single-cell multi-omics to understand chromatin dynamics

Abstract:

The advent of the single-cell transcriptome technology scRNAseq has made determining cell types for tissue organization feasible. Furthermore, acquiring transcriptome data with sufficient data depth has enabled lineage analysis focusing on the similarity of individual cells. On the other hand, elucidating epigenomic regulation at the single cell level committing cell lineages has still been challenging, especially the analysis of transcription factors. We previously developed highly efficient epigenome analysis technologies based on immunostaining, namely chromatin insertion labelling (ChILseq), which enables epigenome analysis at a single-cell level at high depth. We are expanding ChILseq to single-cell multi-omics technology, which can simultaneously analyze individual cells' transcriptome and chromatin state, which is highly anticipated.

In this talk, I would like to introduce our attempt to understand epigenomic regulation by transcription factor at a single cell in skeletal muscle differentiation as a model and other related technologies.
